Like all medicines, Mixtard 50/50 Penfill can cause side effects, although not everybody gets them. **Very Common (may affect more than 1 in 10 people):** * Low blood sugar (hypoglycemia). Early warning signs of low blood sugar may include: cold sweat; cool pale skin; headache; rapid heart beat; feeling sick; feeling very hungry; temporary vision changes; drowsiness; unusual tiredness and weakness; nervousness or tremor; feeling anxious; feeling confused; difficulty in concentrating. **If you experience any of these signs, eat sugary foods or drinks straight away.** **Common (may affect up to 1 in 10 people):** * Local reactions at the injection site: Redness, swelling, and itching at the injection site. * Swelling: Swelling may occur around your ankles and other joints. **Uncommon (may affect up to 1 in 100 people):** * Signs of allergy: Local allergic reactions (redness, swelling, itching) may occur at the injection site. If these spread to other parts of your body, tell your doctor immediately. Serious allergic reactions are very rare, but can be life-threatening. Symptoms of a serious allergic reaction include: swelling of the face, mouth and throat; difficulty breathing; rapid heart beat; skin rash all over your body; feeling faint. * Lipodystrophy: (Pitting in the skin) If you inject insulin too often at the same site, the fatty tissue under the skin at this site may shrink (lipoatrophy) or thicken (lipohypertrophy). Changing the injection site with each injection may help prevent these skin changes. **Very rare (may affect up to 1 in 10,000 people):** * Vision problems: When you first start your insulin treatment, it may disturb your vision, but the reaction usually wears off. * Painful neuropathy: If your blood sugar level improves very fast, you may get nerve related pain – this is usually temporary. * Swollen joints. **Other Side Effects Reported** * Insulin antibodies: Insulin antibodies are produced in the body but usually don't cause any side effects. Very rarely, a high dose of insulin may be required to avoid hyperglycaemia. * Hypoglycemia: If low blood sugar is severe and not treated, it can cause brain damage and death.

Mixtard 50/50 is a biphasic human insulin used to control blood glucose levels in people with diabetes mellitus, usually type 1 or type 2, when insulin therapy is required. It helps manage blood sugar both after meals and between meals or overnight.
Mixtard 50/50 contains 50% soluble (short-acting) insulin and 50% isophane (intermediate-acting) insulin. The soluble part starts lowering blood glucose relatively soon after injection, mainly covering the rise in sugar after meals. The isophane part is absorbed more slowly and provides a longer duration of action, helping to control blood sugar between meals and during the night.
Mixtard 50/50 is injected subcutaneously (under the skin), usually in the abdomen, thigh, buttock, or upper arm. The exact injection technique, dose, and timing must be taught by a healthcare professional. Injection sites should be rotated within the same region to reduce the risk of skin changes such as lipodystrophy. It should not be injected into a vein or muscle unless specifically instructed by a doctor.
Alcohol can affect blood glucose levels and may increase the risk of hypoglycemia when you are using Mixtard 50/50. Drinking on an empty stomach or in large amounts is particularly risky. If you choose to drink alcohol, do so in moderation, avoid skipping meals, monitor your blood glucose more frequently, and be aware of the symptoms of low blood sugar. Discuss alcohol use with your healthcare provider.
Human insulin preparations like Mixtard 50/50 are generally considered safe in pregnancy and are often preferred for blood glucose control in pregnant women with diabetes. However, insulin requirements usually change during pregnancy and after delivery, so close monitoring and dose adjustments by a specialist are essential. Do not start, stop, or change your insulin regimen without medical supervision during pregnancy.
Mixtard 50/50 is generally considered safe to use while breastfeeding. Insulin does not significantly pass into breast milk in a way that harms the baby. However, your insulin needs may change after delivery and during breastfeeding, so your dose may need adjustment. Regular blood glucose monitoring and follow-up with your healthcare provider are important.
People with liver or kidney impairment may have altered insulin requirements and a higher risk of hypoglycemia when using Mixtard 50/50. Dose adjustments and more frequent blood glucose monitoring may be necessary. It is important to inform your doctor about any liver or kidney disease before starting treatment, and to follow their instructions closely. Never change your dose without medical advice.
Mixtard 50/50 itself does not directly impair driving, but low or high blood sugar can affect concentration, reaction time, and judgment. Hypoglycemia is particularly dangerous while driving. Always check your blood glucose before driving, carry a fast-acting carbohydrate snack, and avoid driving if you have symptoms of low blood sugar or if your levels are unstable. Discuss driving safety with your healthcare provider if you have frequent hypoglycemia or reduced awareness of low blood sugar.
If you miss a dose, check your blood glucose and follow the advice given by your healthcare team or diabetes educator. Do not double your next dose to make up for a missed one unless specifically instructed. If you inject too much Mixtard 50/50, you are at risk of hypoglycemia. Monitor your blood sugar closely, take fast-acting carbohydrates if levels are low or if you have symptoms, and seek urgent medical help if severe symptoms such as confusion, seizures, or loss of consciousness occur.
You should not use Mixtard 50/50 if you are allergic to human insulin or any of the excipients in the product. Signs of an allergic reaction may include rash, itching, swelling, difficulty breathing, or severe dizziness. Mild local reactions at the injection site, such as redness or swelling, can occur and often improve with time. If you suspect a serious allergy or if local reactions are severe or persistent, contact your doctor immediately.
The most common side effect of Mixtard 50/50 is hypoglycemia (low blood sugar), which can cause sweating, trembling, hunger, palpitations, headache, irritability, confusion, and in severe cases, loss of consciousness or seizures. Other possible side effects include local injection site reactions (redness, swelling, itching), changes in fat tissue under the skin (lipodystrophy), and rarely, generalized allergic reactions. Report any unusual or severe symptoms to your healthcare provider.
Unopened Mixtard 50/50 should be stored in a refrigerator according to the instructions on the package, and must not be frozen. Once in use, it can usually be kept at room temperature for a limited period as specified by the manufacturer, away from direct heat and light. Always check the product leaflet for exact storage conditions and discard the insulin if it has been improperly stored, is past its expiry date, or appears abnormal.
